Pramiracetam has been synthesised from original racetam. It is thought to be the strongest racetam currently on the market. In comparison to original racetam, it is 30 times more concentrated. Therefore, it is important to take extra care when using it as lower dosages are required (see below). Unlike some other racetams that require a prolonged usage before clear cognitive benefits are observed, Pramiracetam may be effective even after acute dosages.
Pramiracetam enhances acetylcholine neurotransmission in the brain pathways associated with cognitive processes and mental functions, including learning and memory formation. Long-term memory seems to be affected the most. Other cognitive enhancing effects include improved thinking abilities and logic. Some users also report an increase in sensory perceptions. There is very little effect on mood as Pramiracetam does not work by increasing the levels of neurotransmitters that are responsible for it.
Although exact mechanisms of action are not well known, mechanisms underpinning Pramiracetam’s appear to be similar to those of other racetams. Pramiracetam increases the rate of high affinity choline uptake (HACU) in the hippocampus. This leads to a greater acetylcholine synthesis which plays a very important role in the formation and creation of new neuronal connections. In addition, it raises the levels of energy by improving the ATP synthesis. This results in the increase of cAMP levels in the brain. A number of brain receptor sites, including glutamate, are modulated following Pramiracetam use. Interestingly, Pramiracetam has been shown to have promising results in increasing the neural communication between the left and right hemispheres by having a positive effect on the corpus callosum. Such preliminary findings may change the way we learn and improve a range of different skills, such as creativity. It also exhibits a very high oral bioavailability.
The recommended daily dosage is 800 – 1200 mg taken in 2 or 3 equal administrations. Pramiracetam should be taken with a meal. It appears to start working 1-2 hours post-ingestion. Biological half-life of Pramiracetam is 4-6 hours. Therefore timing of acute administration of Pramiracetam before cognitive tasks is very important. Due to its high concentration, Pramiracetam is often used by the experienced nootropics users who have tried other slightly less potent racetams (e.g. Aniracetam and Oxiracetam).
Although Pramiracetam is the strongest racetam there are no adverse side effects associated with its use. However, it is very important to ensure that the recommended dosages are not exceeded. Otherwise some side effects, like headaches and drowsiness, may occur. They can be prevented by simply reducing the dose as well as using it with a choline nootropic.
